A6K-glucose is a novel, self-assembling peptide-drug conjugate designed to treat advanced solid tumors by exploiting the Warburg effect. It consists of a surfactant-like peptide (A6K, typically Ac-AAAAAAK-NH2) conjugated to a glucose moiety. This glucose component allows the molecule to be selectively recognized and internalized by cancer cells that overexpress glucose transporters, such as GLUT1. Once localized at the tumor site or within the cell, the peptide undergoes a structural transition to form stable nanofibers. These nanofibers physically disrupt the cell membrane or interfere with vital cellular functions, leading to selective tumor cell death (necrosis or apoptosis) while sparing healthy tissues with lower glucose demands. It is currently being evaluated in early-phase clinical trials for patients with refractory solid tumors.
