Overview

A72 is a mouse monoclonal antibody targeting human thymidine kinase 1 (TK1), a pyrimidine salvage pathway enzyme involved in DNA synthesis and repair. While TK1 is typically expressed in the cytosol and elevated in the serum of cancer patients, it is also selectively overexpressed on the cell surface of various malignant cells, including breast, lung, colon, and prostate cancers, while remaining absent on normal cells. Developed by researchers at Brigham Young University and licensed to Thunder Biotech, A72 binds to an immunodominant region of TK1 outside of its C-terminal domain. It is being investigated as a potential diagnostic biomarker and therapeutic targeting agent, including for use in antibody-drug conjugates (ADCs) and chimeric antigen receptor (CAR) therapies to selectively eliminate TK1-expressing tumor cells.
