A8 is an immunomodulatory antibody designed to target cell surface B7-H4, a protein whose expression in tumor cells limits CD8 T cell infiltration and creates an immunosuppressive "cold" tumor microenvironment. Developed from a non-immunized human scFv library, A8 aims to overcome primary resistance to anti-PD-1 antibody therapy. It functions by binding to the Ig-like V-type domain of B7-H4, inducing dynamin-dependent endocytosis and lysosomal degradation of B7-H4, which subsequently leads to ferroptosis in B7-H4 positive tumor cell lines. This action promotes immunogenic cell death, dendritic cell maturation, and T cell activation. A8 also enhances T cell cytotoxicity and facilitates the infiltration of CD8+ T cells or HER2 CAR-T cells into tumor sites, thereby inhibiting tumor growth.
