A83-01 is a **potent, selective small molecule inhibitor** of the transforming growth factor-beta (TGF-β) type I receptor kinase (ALK5), activin type IB receptor (ALK4), and nodal type I receptor (ALK7). It blocks TGF-β signaling by inhibiting the phosphorylation of Smad2/3, thereby suppressing downstream effects such as epithelial-to-mesenchymal transition (EMT), apoptosis, and senescence. A83-01 promotes proliferation and maintains the stemness/self-renewal capacity of various cell types in vitro, including human and rat induced pluripotent stem cells and mesenchymal stem cells. It was first described and researched in the context of preventing tumor progression, fibrosis, and enhancing cell culture conditions for stem cell research and regenerative medicine. It is used primarily as a research tool and not as an approved clinical pharmaceutical.
