Overview

AA3-FIBA-BA2 is an experimental small molecule MEK (mitogen-activated protein kinase kinase) inhibitor identified through a high-throughput nanodroplet array platform. It targets the MAPK/ERK (mitogen-activated protein kinase/extracellular signal-regulated kinase) pathway, which is frequently dysregulated in various malignancies, including colorectal and pancreatic cancers. The compound was discovered among a library of 325 MEK inhibitors synthesized using a miniaturized solid-phase combinatorial approach. AA3-FIBA-BA2 demonstrated significantly higher cytotoxicity in cell-based screenings compared to the clinically approved MEK inhibitor mirdametinib. Molecular docking studies indicate that AA3-FIBA-BA2 operates via an allosteric binding mechanism, acting as a non-competitive ATP inhibitor of MEK.
