Overview

AAA503 (also known as NeoB) is a next-generation radiopharmaceutical developed by Advanced Accelerator Applications (a Novartis company). It consists of a DOTA-coupled gastrin-releasing peptide receptor (GRPR) antagonist, NeoBOMB1, labeled with the beta-emitting radioisotope lutetium-177 (177Lu). GRPR is a G-protein coupled receptor that is frequently overexpressed in various solid tumors, including breast cancer, prostate cancer, and gastrointestinal stromal tumors (GIST). Unlike traditional agonists, AAA503 acts as an antagonist, which may offer improved safety and pharmacokinetic profiles by avoiding receptor activation-induced side effects while maintaining high tumor uptake. The drug is designed for peptide receptor radionuclide therapy (PRRT), delivering targeted radiation to destroy cancer cells while sparing healthy tissue.
