Overview

AAB-003 is a humanized monoclonal antibody developed as an immunotherapy for Alzheimer's disease. It is derived from bapineuzumab (AAB-001) and was engineered to reduce Fc-receptor-mediated effector function in order to minimize amyloid-related imaging abnormalities (ARIA), a complication observed with its predecessor. The drug targets amyloid beta-protein, aiming to inhibit its accumulation and promote clearance from the brain. Clinical studies showed dose-dependent increases in plasma amyloid β but no significant changes in cerebrospinal fluid biomarkers. The safety profile indicated that AAB-003 was generally well tolerated up to 8 mg/kg for up to 91 weeks, with ARIA-E being the most notable adverse event at higher doses. Development of AAB-003 for Alzheimer's disease has been discontinued[1][2][3][4][5].
