The AAHI leishmaniasis vaccine is an adjuvant-formulated recombinant protein subunit vaccine program developed by the Access to Advanced Health Institute (formerly the Infectious Disease Research Institute, IDRI). The vaccine candidates, such as Leish-111f and Leish-110f, consist of polyprotein antigens derived from various *Leishmania* species. These antigens are formulated with proprietary Toll-like receptor 4 (TLR4) agonist adjuvants, most notably GLA-SE (Glucopyranosyl Lipid Adjuvant in a Stable Emulsion) or MPL-SE. The primary mechanism of action is the induction of a potent Th1-type cellular immune response, characterized by the production of interferon-gamma (IFN-γ) by CD4+ T cells. This response is critical for activating macrophages to eliminate intracellular *Leishmania* parasites. The program targets multiple forms of the disease, including visceral leishmaniasis (VL), cutaneous leishmaniasis (CL), and post-kala-azar dermal leishmaniasis (PKDL), for both prophylactic and therapeutic applications.
