AAHI-SC2 is a self-amplifying RNA (saRNA) vaccine candidate designed to prevent COVID-19 by targeting the SARS-CoV-2 spike (S) protein. Developed by the Access to Advanced Health Institute (AAHI) in collaboration with ImmunityBio and AccessBio, the vaccine utilizes a nanostructured lipid carrier (NLC) delivery system that incorporates an adjuvant, making it both self-amplifying and self-adjuvanted. A significant advantage of AAHI-SC2 is its thermostability; it can be lyophilized for storage at standard refrigerated temperatures for at least 10 months or at room temperature for up to six months, facilitating distribution in resource-limited settings. It is currently being evaluated as a booster in the Phase 1/2 THEMBA II clinical trial (NCT05370040) in South Africa.
