AAHI-SC3 is a self-amplifying RNA (saRNA) vaccine candidate developed by the Access to Advanced Health Institute (AAHI) in collaboration with ImmunityBio for the prevention of COVID-19. Unlike conventional mRNA vaccines, saRNA vaccines incorporate genetic machinery derived from alphaviruses that allows the RNA to replicate itself once inside the host cell. This self-amplification process potentially enables lower dosing requirements and more durable immune responses compared to standard mRNA platforms. AAHI-SC3 is formulated using AAHI's proprietary Lipid Inorganic Nanoparticle (LION) delivery system, which is designed to enhance the stability of the RNA and act as an adjuvant to boost immunogenicity. The vaccine is currently being evaluated as a booster in the THEMBA II Phase 1/2 clinical trial in South Africa, targeting adults who have been previously vaccinated or infected with SARS-CoV-2.
