AAN-14a is a preclinical-stage in vivo CAR-T cell therapy being developed by Aanastra in collaboration with DIVINCELL. The therapy utilizes DIVINCELL's proprietary PEP-NP™ (peptide-based nanoparticle) delivery platform, also referred to as PEP-CAR™, to deliver anti-CD19 CAR mRNA directly to T cells within the patient's body. By targeting CD3/CD5+ T cells, the nanoparticles program these cells to express a chimeric antigen receptor (CAR) that engages CD19 on B cells, leading to their depletion. This approach is designed to treat hematological malignancies and autoimmune diseases while bypassing the logistical challenges and toxicities associated with traditional ex vivo CAR-T manufacturing and viral or lipid nanoparticle delivery systems. Preclinical studies have demonstrated the potential for safe, repeated dosing without significant immunogenicity or liver toxicity.
