AAV(DJ)-Fosb-ZFP-p65

AAV(DJ)-Fosb-ZFP-p65 is a preclinical gene therapy construct designed to selectively regulate the expression of the endogenous *FosB* gene in specific brain regions, such as the nucleus accumbens (NAc). It consists of an adeno-associated virus serotype DJ (AAV-DJ) vector delivering a targeted zinc finger protein (ZFP) fused to the p65 activation domain of the NF-κB transcription factor. The ZFP is engineered to bind specifically to the promoter region of the *FosB* gene, while the p65 domain recruits histone acetyltransferases (such as p300/CBP) to promote histone H3K9/14 acetylation, thereby upregulating endogenous *FosB* (and its isoform ΔFosB) expression. This epigenetic editing tool is primarily investigated in preclinical models of chronic social defeat stress (CSDS), sleep architecture alterations, diurnal rhythm disruptions, and drug addiction to study the role of cell-type-specific *FosB* expression in mediating stress resilience or susceptibility.
