Overview

AAV-CHRDL2 knockdown is an experimental gene therapy agent designed to suppress the expression of Chordin-like 2 (CHRDL2) using an adeno-associated virus (AAV) vector. In the context of inflammatory bowel disease (IBD) research, this tool is utilized to perform loss-of-function studies in vivo. Research presented at ECCO 2026 demonstrated that knocking down CHRDL2 in mouse models of DSS-induced colitis exacerbates intestinal inflammation and impairs the repair of the mucosal barrier. This effect occurs because CHRDL2 normally acts as a bone morphogenetic protein (BMP) antagonist that activates the EGFR-MUC5B/β-catenin signaling pathway to promote intestinal stem cell (ISC) proliferation and regeneration. Consequently, AAV-CHRDL2 knockdown serves as a critical research tool for validating the CHRDL2-EGFR axis as a therapeutic target for intestinal epithelial repair.
