Drug intelligence / Profile preview
AAV-Gna13CA is an adeno-associated virus (AAV)-based gene therapy designed to locally overexpress a constitutively active form of the guanine nucleotide-binding protein subunit alpha-13 (Gα13, encoded by the Gna13 gene). Developed by researchers at Tulane University, AAV-Gna13CA is being investigated for the treatment of rheumatoid arthritis (RA). Gα13 acts as a master negative endogenous regulator of macrophage-mediated inflammation and osteoclastogenesis. By delivering the constitutively active Gna13CA, the therapy suppresses the Gα13-TLR-NF-κB signaling pathway, thereby reducing synovial inflammation, immune cell infiltration, and pathological bone and cartilage destruction in preclinical models of rheumatoid arthritis.
