Drug intelligence / Profile preview
AAV-MEDI4893 is a preclinical gene therapy candidate designed for vectored immunoprophylaxis (VIP) against *Staphylococcus aureus* infections. Developed by researchers at the University of Calgary, University of Guelph, Western University, and Avamab Pharma, it utilizes an adeno-associated virus (AAV) vector (specifically the muscle-tropic AAV6.2FF capsid) to deliver the gene encoding MEDI4893 (suvratoxumab), a humanized monoclonal antibody. Following intramuscular administration, the vector drives sustained, long-term in vivo expression and secretion of MEDI4893. The expressed antibody binds to and neutralizes *S. aureus* alpha-toxin (AT), a key virulence factor that causes cellular lysis, platelet aggregation, and microvascular injury. This approach aims to provide durable prophylactic protection against severe staphylococcal diseases, such as pneumonia and skin infections, without the need for repeated antibody administrations.
