AAV-miR-29b is an adeno-associated virus (AAV) vector-based gene therapy designed to deliver microRNA-29b (miR-29b) to the peritoneal cavity. Developed by researchers at Jichi Medical University, it is intended for the treatment and prevention of peritoneal metastasis (PM) in gastrointestinal cancers, including gastric and pancreatic cancer. The therapy works by transducing peritoneal mesothelial cells (PMCs), leading to the secretion of miR-29b-enriched extracellular vesicles. This process suppresses mesothelial mesenchymal transition (MMT) and peritoneal fibrosis, which are critical for the attachment and growth of metastatic tumor cells. In preclinical syngeneic murine models, a single intraperitoneal injection of AAV-miR-29b significantly reduced the number of metastatic nodules and demonstrated synergistic effects when combined with low-dose paclitaxel.
