Overview

AAV-PIK3CA H1047R is an adeno-associated virus (AAV) vector-based gene therapy construct designed to induce the somatic activation of the PIK3CA H1047R mutation. This specific mutation is a common oncogenic driver in human breast cancers, particularly in estrogen receptor-positive (ER+) subtypes, leading to constitutive activation of the PI3K/AKT/mTOR signaling pathway. In preclinical research, this vector is delivered intraductally into rat mammary glands to model the evolution of breast cancer, specifically ductal and invasive lobular carcinomas (ILC). While AAV-PIK3CA H1047R can independently drive tumor formation with a latency of approximately four months, it is frequently utilized in combination with other genetic modifications, such as CDH1 (E-cadherin) knockout, to create more clinically relevant models of hormone-dependent invasive lobular breast cancer.
