Drug intelligence / Profile preview
AAV-TBG-KCNJ2 is an experimental adeno-associated virus (AAV) gene therapy vector designed to deliver the potassium inwardly rectifying channel subfamily J member 2 (KCNJ2) gene specifically to hepatocytes [1]. Driven by the liver-specific thyroxine-binding globulin (TBG) promoter, this construct overexpresses the Kir2.1 inward rectifying potassium channel to induce chronic hyperpolarization of hepatocytes [1]. In preclinical models, AAV-TBG-KCNJ2 is utilized to investigate the physiological impacts of hepatocyte membrane potential on hepatic vagal afferent nerve firing, physical activity, and systemic energy expenditure, offering insights into potential therapeutic targets for obesity and metabolic homeostasis [1].
