AAV2-shChi3l1 is an adeno-associated virus serotype 2 (AAV2) vector-based gene therapy designed to deliver short hairpin RNA (shRNA) targeting Chitinase-3-like protein 1 (CHI3L1). CHI3L1, also known as YKL-40, is a secreted glycoprotein that plays a significant role in inflammation, tissue remodeling, and cancer progression. In the context of hepatocellular carcinoma (HCC), CHI3L1 is induced in pulmonary epithelial cells by tumor-derived exosomes, facilitating the formation of a pre-metastatic niche in the lungs by promoting angiogenesis and cell migration. AAV2-shChi3l1 mediates the knockdown of CHI3L1 expression, thereby disrupting the establishment of this niche and significantly reducing pulmonary metastasis.
