Overview

AAV6-CXCL9 is a recombinant adeno-associated virus (AAV) gene therapy designed to modulate the tumor microenvironment (TME) of glioblastoma (GBM). It utilizes an AAV6 vector to deliver the gene for C-X-C motif ligand 9 (CXCL9), a potent chemokine that recruits CXCR3-positive cytotoxic T lymphocytes (CTLs) and natural killer T (NKT) cells. In glioblastoma, CXCL9 is typically absent, leading to a cold tumor microenvironment devoid of lymphocytes. By locally expressing CXCL9, primarily in peri-tumoral astrocytes, the therapy aims to increase lymphocyte infiltration and sensitize the tumor to immune checkpoint inhibitors like anti-PD-1 antibodies. This approach is being developed by researchers at the University of Florida and Emory University.
