The **AAV6 multivalent melanoma vaccine** is a preclinical viral-vector cancer vaccine developed in an academic setting for melanoma. It combines capsid- and expression cassette-optimized adeno-associated virus serotype 6 vectors encoding four melanoma-associated self-antigens: premelanosome protein gp100, tyrosinase, tyrosinase-related protein 1, and dopachrome tautomerase. The modified AAV6-S663V capsid is intended to improve intracellular trafficking and antigen presentation after intramuscular administration, producing antigen-specific cellular and humoral immunity. In murine B16F10 melanoma models, the vaccine approach reduced metastatic lung tumor burden and delayed solid-tumor growth, including in combination with PD-1 and PD-L1 checkpoint blockade.
