Drug intelligence / Profile preview
AAV8-adiponectin is a preclinical gene therapy candidate developed by AstraZeneca for the treatment of metabolic and renal disorders, including diabetic kidney disease (DKD), dyslipidemia, and obesity. The therapeutic agent consists of an adeno-associated virus serotype 8 (AAV8) vector engineered with an adipocyte-specific promoter to drive the in vivo overexpression of adiponectin, an endocrine factor synthesized and released from adipose tissue. By targeting adipose tissue and promoting the sustained systemic exposure of functional adiponectin, the therapy aims to mitigate metabolic dysfunction and protect against kidney damage. In preclinical models of obesity and diabetic kidney disease, treatment with AAV8-adiponectin has demonstrated the ability to reduce body weight gain, improve hepatic metabolic parameters (such as decreasing liver triglycerides and cholesterol), and transiently lower the urinary albumin-to-creatinine ratio (UACR).
