AAV9-luciferase is a preclinical research tool consisting of an adeno-associated virus serotype 9 (AAV9) vector engineered to express the firefly luciferase reporter protein. It is primarily used in bioluminescence imaging (BLI) to monitor gene delivery efficiency, tissue-specific transduction, and the long-term biodistribution of AAV9-based gene therapies in vivo. By measuring the light emitted upon the administration of a luciferin substrate, researchers can non-invasively track the expression of the transgene across various tissues, including the liver, heart, skeletal muscle, and central nervous system. While not a therapeutic agent, it serves as a critical benchmark for optimizing vector dosing and evaluating potential toxicities, such as pulmonary endothelial expression or cytokine activation, in animal models of diseases like Duchenne muscular dystrophy (DMD).
