Overview

AAV9-NLRP6 is an experimental gene therapy construct utilizing an adeno-associated virus serotype 9 (AAV9) vector to deliver and overexpress the NLRP6 (NLR Family Pyrin Domain Containing 6) gene. NLRP6 is a member of the nucleotide-binding oligomerization domain-like receptor family that plays a pivotal role in maintaining intestinal homeostasis, regulating the gut microbiota, and modulating inflammatory responses. In the context of aging and sepsis, research indicates that gut dysbiosis—specifically an increase in *Klebsiella aerogenes*—leads to elevated histamine production, which inhibits NLRP6 expression. This suppression impairs the binding of NLRP6 to LC3, thereby disrupting autophagy and compromising the intestinal barrier. AAV9-NLRP6 is designed to restore NLRP6 levels, facilitating the NLRP6-LC3-mediated autophagy pathway to repair the gut barrier and ameliorate systemic inflammation in septic conditions.
