AAV9-tMCK-CaMKIICA is an adeno-associated virus (AAV) serotype 9-based gene therapy vector designed to deliver a constitutively active form of Calcium/calmodulin-dependent protein kinase II (CaMKII) specifically to skeletal muscle. It utilizes the truncated muscle creatine kinase (tMCK) promoter to ensure muscle-restricted transgene expression. This construct is primarily used as a research tool to investigate the role of chronic CaMKII activation in muscle pathology. Studies using this vector have demonstrated that persistent CaMKII signaling drives features of sarcopenia, including muscle atrophy, mitochondrial dysfunction, and impaired contractile performance, thereby identifying CaMKII as a potential therapeutic target for age-related muscle decline.
