AAVLP-HPV is a novel virus-like particle (VLP) vaccine targeting human papillomavirus (HPV), developed by 2A Pharma. It is the world’s only L2-based HPV vaccine in clinical testing and aims to provide significantly broader protection than currently approved HPV vaccines by targeting the minor capsid protein L2 rather than the major capsid protein L1. The vaccine uses adeno-associated virus-like particles (AAVLPs) as a platform to display neutralizing epitopes from multiple HPV types, including key regions of the L2 protein from different strains such as HPV16 and HPV31. Preclinical studies have shown that this approach induces high titers of cross-neutralizing antibodies against several oncogenic and non-oncogenic HPV types. The first-in-human phase 1 trial demonstrated that AAVLP-HPV was safe, well tolerated, and elicited robust antibody responses against multiple L2 epitopes in healthy adults[4][5][6][8]. The product is being developed for prevention of cervical, skin, oropharyngeal, and other cancers related to persistent infection with various types of human papillomavirus.
