AB-12 is an intravenous small molecule autophagy-targeting chimera (AUTOTAC) being developed by Autotac Bio for the treatment of Alzheimer's disease. It is designed to selectively induce the degradation of pathological Tau protein aggregates, a key hallmark of Alzheimer's disease, by recruiting the p62/SQSTM1 autophagy receptor. This recruitment facilitates the sequestration of Tau into autophagosomes for subsequent degradation via the autophagy-lysosome pathway. Unlike traditional proteolysis-targeting chimeras (PROTACs) that utilize the ubiquitin-proteasome system, the AUTOTAC platform enables the clearance of larger protein aggregates and potentially other cellular debris. AB-12 is also being investigated in an oral formulation.
