AB-19 (also identified as ATB2005A) is an orally bioavailable small molecule Autophagy-Targeting Chimera (AUTOTAC) developed by Autotac Bio for the treatment of progressive supranuclear palsy (PSP) and other tauopathies. The compound is a bifunctional chimeric molecule designed to simultaneously bind pathological, β-sheet-rich tau aggregates and the autophagic receptor p62 (SQSTM1). By recruiting these aggregates to the p62 receptor, AB-19 facilitates their sequestration into autophagosomes and subsequent degradation via the lysosomal pathway. This mechanism of action specifically targets neurotoxic tau aggregates for clearance while sparing monomeric tau. Preclinical studies in mouse models have demonstrated that the drug can reduce intraneuronal tau aggregates and improve neuroinflammation, cognitive function, and motor behavior. A Phase 1 clinical trial in healthy adults was initiated in early 2025 to evaluate the safety, tolerability, and pharmacokinetics of the compound.
