AB-201 is an investigational allogeneic chimeric antigen receptor natural killer (CAR-NK) cell therapy targeting HER2 (ErbB-2), developed for the treatment of HER2-positive solid tumors, including breast cancer and gastric/gastroesophageal junction adenocarcinoma. The therapy uses umbilical cord blood-derived NK cells engineered to express a high-affinity anti-HER2 single-chain variable fragment (scFv), coupled with a unique costimulatory structure and IL-15 expression for enhanced activity and persistence. The underlying NK cells are preselected for advantageous attributes such as high-affinity CD16 receptor variants and KIR-B haplotype. AB-201 is designed for large-scale manufacturing using Artiva’s AlloNK platform, enabling cryopreserved, infusion-ready product suitable for repeat outpatient dosing. Preclinical studies have demonstrated potent anti-tumor activity in multiple HER2-positive tumor models[1][5][6][7].
