AB-2100 is an investigational autologous cellular immunotherapy developed for the treatment of advanced or metastatic clear-cell renal cell carcinoma (ccRCC). It is a next-generation chimeric antigen receptor (CAR) T-cell therapy engineered using Arsenal Biosciences' CITE (CRISPR Integration of Transgenes by Electroporation) technology. The patient's own T cells are collected and genetically modified via precise CRISPR-mediated insertion of a synthetic DNA cassette into Chromosome 11. This cassette encodes a synthetic logic gate that requires the presence of two distinct proteins—prostate-specific membrane antigen (PSMA) on tumor endothelium and carbonic anhydrase 9 (CA9) on adjacent tumor cells—for activation, thereby increasing selectivity for ccRCC tumors while sparing normal tissues. Additional features include shRNAs to protect the engineered T cells from immunosuppressive signals in the tumor microenvironment and a synthetic pathway activator to enhance potency and persistence. AB-2100 is administered intravenously following conditioning chemotherapy[1][2][3][4][5][6][8].
