Overview

AB-3028 is a next-generation, programmable circuit CAR T cell therapy being developed by ArsenalBio for the treatment of metastatic castration-resistant prostate cancer (mCRPC). It is described as a dual-targeting, sequential AND logic–gated CAR T product that uses one antigen (a “PrimeR” target) to license or prime the T cells and a second established CAR target to drive full activation, thereby enhancing tumor selectivity and reducing recognition of normal tissues.[1][5] AB-3028 incorporates additional engineering features to improve T-cell fitness in the suppressive tumor microenvironment, including modules designed to enhance persistence, limit exhaustion after repeated antigen encounters, and counteract TGF-β–mediated immunosuppression.[1][3] Preclinical studies have shown anti-tumor efficacy and survival benefit in prostate cancer xenograft models at doses lower than earlier CAR constructs, supporting advancement into first-in-human trials for mCRPC.[1][3][8]
