Overview

AB-33 is an autophagy-targeting chimera (AUTOTAC) small molecule degrader being developed by Autotac Bio for the treatment of pancreatic cancer. It is designed to selectively target and induce the degradation of the p53 protein, which is frequently mutated and serves as a key oncogenic driver in pancreatic ductal adenocarcinoma. The AUTOTAC platform utilizes bifunctional molecules that recruit the target protein to the autophagy adapter protein p62 (SQSTM1). This recruitment facilitates the sequestration of the target protein into autophagosomes, leading to its subsequent degradation via the lysosomal pathway. This approach offers a novel therapeutic strategy for eliminating proteins that are traditionally considered undruggable or those that have acquired gain-of-function mutations.
