Drug intelligence / Profile preview
Ab-ECSnps is a dopamine receptor D3 (DRD3) antibody-conjugated, 17β-estradiol (E2)-loaded chitosan nanoparticle formulation developed by the Institute of Nano Science and Technology (INST), Mohali. It is designed for targeted delivery of 17β-estradiol to the brain to treat Parkinson's disease. By targeting DRD3 on dopaminergic neurons, the nanoparticles deliver E2 directly to the brain, minimizing peripheral side effects. E2 acts as a specific calpain inhibitor, preventing calpain translocation into mitochondria and promoting the mitochondrial translocation of the epigenetic regulator BMI1 (a Polycomb repressive complex 1 member). This dual action preserves mitochondrial homeostasis, enhances cell viability, increases mitochondrial DNA copy number, and maintains mitochondrial membrane potential. Additionally, Ab-ECSnps activates SIRT1-dependent autophagy and inhibits caspase-3-driven apoptosis, protecting dopaminergic neurons from toxicity in preclinical models of Parkinson's disease.
