Overview

AB-TNF is an oral small-molecule inhibitor of Tumor Necrosis Factor (TNF) currently in preclinical development by Arcus Biosciences in collaboration with Gilead Sciences. Unlike existing anti-TNF biologics that non-selectively block TNF activity, AB-TNF is engineered to selectively inhibit TNFR1 signaling while preserving TNFR2 signaling. This selective mechanism is intended to block the pro-inflammatory pathways associated with TNFR1 while maintaining the immune-regulatory and neuroprotective functions of TNFR2, potentially offering an improved safety profile and enhanced efficacy. The program targets chronic inflammatory conditions, including rheumatoid arthritis, psoriasis, and inflammatory bowel disease, with an Investigational New Drug (IND) application anticipated in late 2026 or early 2027.
