**Ab-TP-MDNP** is an investigational, intravenously administered brain-penetrating manganese dioxide-polymer-lipid hybrid nanomedicine developed at the University of Toronto for Alzheimer’s disease. It comprises a blood-brain barrier-crossing terpolymer-containing manganese dioxide nanoparticle functionalized with the anti-amyloid beta antibody clone 4G8. The anti-amyloid beta component localizes the construct to amyloid beta plaques and oligomers, while manganese dioxide reacts with local reactive oxygen species, including hydrogen peroxide, to reduce oxidative stress and generate oxygen and paramagnetic manganese ions. In transgenic Alzheimer’s disease mouse models, the construct has been reported to reduce hypoxia, neuroinflammation, vascular dysfunction, amyloid beta pathology, and cognitive impairment; it has also supported MRI-based detection of early neuroinflammation.
