Ab.20-vcMMAE is a preclinical antibody-drug conjugate (ADC) developed by Biocytogen. It consists of a fully human monoclonal antibody (Ab.20) targeting Cadherin-3 (CDH3/P-cadherin) conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a cathepsin-cleavable valine-citrulline (vc) linker. CDH3 is a cell adhesion molecule frequently overexpressed in several malignant solid tumors, including pancreatic, breast, lung, and colorectal cancers, while showing limited expression in normal tissues. Ab.20-vcMMAE was utilized as a proof-of-concept benchmark to demonstrate the efficacy of CDH3-targeted delivery, showing robust anti-tumor activity in patient-derived xenograft (PDX) models. These findings supported the subsequent development of BCG014, which utilizes the same antibody clone with a novel topoisomerase I inhibitor payload.
