AB011 + capecitabine + oxaliplatin is an investigational combination regimen developed for the treatment of advanced gastric cancer and gastroesophageal junction (GEJ) adenocarcinoma. The combination consists of AB011, a recombinant humanized IgG1 monoclonal antibody targeting Claudin18.2 (CLDN18.2), capecitabine (an oral fluoropyrimidine prodrug that is converted to 5-fluorouracil in vivo and inhibits DNA synthesis), and oxaliplatin (a platinum-based chemotherapeutic that induces DNA cross-linking and apoptosis). AB011 enhances antitumor efficacy via immune-mediated mechanisms of action directed at CLDN18.2, which is overexpressed in various solid tumors, particularly gastric cancers. Preclinical and clinical data demonstrate synergistic effects between AB011 and cytotoxic agents, with the combination showing early clinical benefit, manageable safety, and high objective response rates in Phase I studies.
