Drug intelligence / Profile preview
AB204 (also known as DRT-102) is a recombinant Activin A/BMP-2 chimeric fusion protein designed as a bone morphogenetic protein (BMP) super-agonist for bone regeneration and spinal fusion. Developed by Darnatein (a subsidiary of OSR Holdings), AB204 was engineered using a design-augmented synthetic biology platform to combine the osteoinductive potency of BMP-2 with the receptor-binding properties of Activin A. This chimeric structure confers resistance to the natural BMP antagonist Noggin and significantly increases its affinity for the type II receptor ActRII and type I receptor BMPR1A compared to wild-type BMP-2. Preclinical and early clinical studies in South Korea have demonstrated that AB204 promotes robust osteogenesis and spinal bone fusion at substantially lower doses than recombinant human BMP-2 (rhBMP-2), potentially offering a safer alternative with reduced risk of adverse effects such as ectopic bone formation, inflammation, or tumor progression.
