Overview

AB569 is a novel, non-antibiotic, bactericidal combination drug composed of acidified nitrite (A-NO2−) and ethylenediaminetetraacetic acid (EDTA). It is designed to eradicate multidrug-resistant Gram-negative and Gram-positive bacteria, including pathogens such as Pseudomonas aeruginosa and Acinetobacter baumannii. The mechanism of action involves the generation of bactericidal levels of nitric oxide from acidified nitrite, while EDTA chelates divalent cations destabilizing bacterial membranes and stabilizes S-nitrosyl thiols, enhancing the antimicrobial effect. This dual action leads to catastrophic loss of bacterial core biosynthetic pathways (DNA, RNA, protein synthesis), iron metabolism disruption, and biofilm eradication. Preclinical studies have shown efficacy in killing bacteria in vitro and in animal models without toxicity to human cells at therapeutic concentrations. AB569 has been developed for use as an inhaled therapy for chronic lung infections (notably cystic fibrosis [CF] and chronic obstructive pulmonary disease [COPD]), as well as a topical agent for wound healing—including burn injuries—where it also promotes tissue repair.
