Ab612-ADC is an antibody-drug conjugate (ADC) targeting CD171 (L1 cell adhesion molecule, L1CAM), a cell-adhesion molecule implicated in epithelial-mesenchymal transition (EMT), receptor tyrosine kinase activation, and neuroendocrine differentiation in various solid tumors. Developed by APITBIO in collaboration with AbTis, the ADC utilizes the novel anti-CD171 monoclonal antibody Ab612. This antibody is conjugated to either monomethyl auristatin E (MMAE) with a drug-to-antibody ratio (DAR) of 2 or exatecan with a DAR of 4, using AbTis's proprietary Abclick® Pro linker. The linker is designed to preserve antibody-FcRn interactions, potentially extending the drug's half-life. Preclinical data presented at AACR 2026 showed that Ab612-ADCs exhibit high binding affinity in acidic tumor microenvironments and demonstrate potent antitumor activity in resistant cancer models, including trastuzumab-resistant breast cancer and TKI-resistant lung cancer.
