Overview

Ab7326 (also known as UCB6114) is a high-affinity, allosteric monoclonal antibody targeting Gremlin-1 (GREM1), a secreted glycoprotein and Bone Morphogenetic Protein (BMP) antagonist. Gremlin-1 is frequently overexpressed in the stroma of various solid tumors and in the bone marrow microenvironment of hematological malignancies, where it promotes tumor cell proliferation and stemness by inhibiting BMP-2, 4, and 7 signaling. Ab7326 binds to a novel epitope on Gremlin-1 that is distal to the BMP-binding site, acting as an allosteric inhibitor to restore BMP pathway activity (evidenced by SMAD1/5/8 phosphorylation). Developed by UCB Biopharma, Ab7326 has shown significant preclinical efficacy in reducing tumor burden and metastasis in models of colorectal cancer, multiple myeloma, breast cancer, and prostate cancer.
