Overview

This is a combination antiretroviral therapy (ART) regimen used in the treatment of HIV-1 infection. It consists of four drugs: - **Abacavir** and **lamivudine**, both nucleoside reverse transcriptase inhibitors (NRTIs), which inhibit HIV reverse transcriptase by competing with natural substrates and causing chain termination during viral DNA synthesis. - **Darunavir**, a protease inhibitor, prevents the cleavage of HIV polyproteins, thereby inhibiting viral maturation. - **Ritonavir** acts as a pharmacokinetic enhancer ("booster") for darunavir by inhibiting cytochrome P450 3A4 (CYP3A4), increasing darunavir plasma concentrations. This regimen is considered effective and well-tolerated in both treatment-naïve and experienced patients, achieving high rates of virological suppression[1][3]. It is not available as a fixed-dose co-formulation but can be prescribed as individual components taken together. The combination is particularly used when tenofovir-based regimens are unsuitable or contraindicated[1][5].
