Overview

Abaloparatide is a synthetic 34-amino acid peptide analog of parathyroid hormone-related protein (PTHrP), developed as an anabolic agent for the treatment of osteoporosis. It acts as a selective agonist at the parathyroid hormone 1 receptor (PTH1R), stimulating bone formation by activating the cAMP signaling pathway in osteoblasts and osteocytes. Abaloparatide is indicated for use in postmenopausal women and men with osteoporosis who are at high risk for fracture or have failed or are intolerant to other osteoporosis therapies. The drug has demonstrated significant reductions in vertebral and nonvertebral fractures, with a lower incidence of hypercalcemia compared to teriparatide. The most common side effects include hypercalciuria, dizziness, nausea, headache, palpitations, fatigue, upper abdominal pain, and vertigo[1][3][4][5][6].
