Drug intelligence / Profile preview
Abaloparatide-sMTS is a **microstructured transdermal system** formulation of abaloparatide, a synthetic peptide analog of parathyroid hormone-related protein (PTHrP 1-34) developed for the treatment of osteoporosis in postmenopausal women at high risk for fracture. This formulation uses an array of microneedles coated with the drug to deliver abaloparatide through the skin, providing an alternative to daily subcutaneous injections. The mechanism of action is activation of the parathyroid hormone 1 receptor, stimulating new bone formation and increasing bone mineral density (BMD). Clinical trials show that abaloparatide-sMTS provides consistent pharmacokinetics, significant increases in BMD, and high patient satisfaction, though noninferiority to subcutaneous abaloparatide for lumbar spine BMD increase was not established. The most common adverse events are application site reactions (erythema, pain, swelling), mostly mild or moderate. The product is currently in phase 3 clinical trials for osteoporosis in postmenopausal women[1][3].
