Overview

This is a combination of three biologic disease-modifying antirheumatic drugs (DMARDs): abatacept, adalimumab, and rituximab. Each drug targets different components of the immune system involved in autoimmune diseases such as rheumatoid arthritis. Abatacept is a fusion protein that inhibits T-cell activation by blocking the CD80/CD86-CD28 costimulatory signal. Adalimumab is a monoclonal antibody that binds to tumor necrosis factor alpha (TNF-α), inhibiting its pro-inflammatory effects. Rituximab is a chimeric monoclonal antibody targeting CD20 on B cells, leading to their depletion. While each agent has demonstrated efficacy individually or in combination with non-biologic DMARDs like methotrexate, combining multiple biologics such as these three together is not standard practice due to increased risk of serious adverse events, particularly infections[2][7]. This combination would be considered experimental and potentially unsafe outside of clinical trials.
