Overview

CD20, also known as MS4A1, is a non-glycosylated phosphoprotein expressed on the surface of B lymphocytes from the pre-B cell stage through mature B cells. It is a key regulator of B-cell development, differentiation, and BCR signaling. As a therapeutic target, it is engaged by monoclonal antibodies like rituximab to deplete B cells in B-cell malignancies and autoimmune diseases. CD20 does not internalize or shed upon antibody binding, making it an effective target for complement-mediated cytotoxicity.

Mechanism of action

Antibody binding induces clustering/capping of CD20, promoting complement-dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC), leading to B-cell depletion.
