Overview

Abatacept + aldesleukin is an investigational combination therapy consisting of abatacept, a selective T-cell co-stimulation modulator, and aldesleukin (recombinant interleukin-2). Abatacept is a fusion protein that inhibits T-cell activation by binding to CD80 and CD86 on antigen-presenting cells, thereby blocking the interaction with CD28 on T cells. Aldesleukin is a recombinant form of human interleukin-2 (IL-2) that stimulates the proliferation and activation of regulatory T cells (Tregs) as well as other immune cell subsets. The rationale for combining these agents is to enhance endogenous regulatory T cell numbers and function while suppressing pathogenic immune responses. This combination has been studied in early-phase clinical trials for amyotrophic lateral sclerosis (ALS), aiming to determine safety, tolerability, and effects on regulatory T cell populations[3][5].
