Overview

ABBV-011 is an investigational antibody-drug conjugate (ADC) developed for the treatment of small cell lung cancer (SCLC). It consists of a humanized, cysteine-engineered IgG1 monoclonal antibody targeting seizure-related homolog protein 6 (SEZ6), which is overexpressed in SCLC. The antibody is conjugated via a novel noncleavable linker to the cytotoxic payload N-acetyl-gamma-calicheamicin, a potent DNA-damaging agent. Upon binding to SEZ6-expressing tumor cells, ABBV-011 is internalized and releases calicheamicin intracellularly, inducing cell death. Preclinical studies demonstrated dose-dependent antitumor activity in SEZ6-positive models, and early-phase clinical trials have shown promising efficacy and tolerability in heavily pretreated patients with relapsed or refractory SCLC[1][2][3][4][5][6].
