ABBV-1042

ABBV-1042 is an investigational orally administered small molecule antagonist of Toll-like receptor 7 (TLR7) and Toll-like receptor 8 (TLR8), developed by AbbVie. These endosomal receptors are key components of the innate immune system that recognize single-stranded RNA; their overactivation is strongly linked to the production of pro-inflammatory cytokines and the pathogenesis of autoimmune diseases, such as systemic lupus erythematosus (SLE). By inhibiting both TLR7 and TLR8, ABBV-1042 aims to modulate pathological immune signaling and reduce the inflammatory cascade. As of late 2024, the drug is in early-stage clinical development, with first-in-human Phase 1 trials evaluating its safety, tolerability, and pharmacokinetics in healthy volunteers.
