ABBV-154

**ABBV-154** is an investigational immunology antibody-drug conjugate developed by AbbVie. It consists of the anti-tumor necrosis factor antibody adalimumab covalently linked through a short hydrophilic linker to a phosphorylated proprietary glucocorticoid receptor modulator payload, A-1677770. The adalimumab component binds TNF, including transmembrane TNF on activated immune cells, while the payload is intended to provide localized glucocorticoid receptor agonism and anti-inflammatory activity with reduced systemic glucocorticoid exposure. ABBV-154 was evaluated in phase 2 studies in rheumatoid arthritis, Crohn's disease, and glucocorticoid-dependent polymyalgia rheumatica, but AbbVie voluntarily ended all clinical development effective April 27, 2023 because its benefit-risk profile was not sufficiently differentiated from available therapies.
